Activities of cell wall hydrolases of Euphorbia heterophylla L. (wild poinsettia) endosperm were investigated during pre- and post-emergence periods, defined as the time interval before and after 2.2 days from the start of imbibition, respectively. The activities of endo-beta-mannanase and beta-mannosidase are higher over the pre-emergence when compared to the post-emergence period and they may be involved in the process of germination in E. heterophylla. Compositional changes of proteins and amino acids in germinating coffee seeds BABT

Endosperm is the main reserve tissue in coffee seeds. Coffee (Coffea arabica L.) seeds were germinated for six weeks and qualitative and quantitative changes in amino acids and proteins were investigated. The total content of free amino acids were reduced during germination, however, protein content remained constant. SDS-PAGE profiles showed that legumin-like proteins became less stained in the last weeks. Asparagine, glutamic acid, aspartic acid, alanine and lysine were the major free amino acids, although serine and glutamine were also significant. In commercial coffee species (Coffea arabica and Coffea canephora), fruit development is a lengthy process, characterized by tissue changes and evolutions. For example, soon after fecundation and up to mid development, the fruit is mainly constituted of the pericarp and perisperm tissue. Thereafter, the perisperm gradually disappears and is progressively replaced by the endosperm (true seed). Initially present in a "liquid" state, the endosperm hardens as it ripens during the maturation phase, as a result of accumulation of storage proteins, sucrose and complex polysaccharides representing the main reserves of the seed. Influence of air temperature on proteinase activity and beverage quality in Coffea arabica Rev. Bras. Bot.

Fruits were collected from trees of Coffea arabica cv. Obatã grown at Mococa and Adamantina in São Paulo State, Brazil, which are regions with marked differences in air temperature that produce coffee with distinct qualities. Mococa is a cooler location that produces high-quality coffee, whereas coffee from Adamantina is of lower quality. The amino acid and protein contents, amino acid profile, and proteinase activity and type in endosperm protein extracts were analysed. Proteinase genes were identified, and their expression was assayed. All results indicate that temperature plays a role in controlling proteinase activity in coffee endosperm. Emergence and stand establishment of tomato (Lycopersicon lycopersicum (L.) (Karsten ex Farw) and pepper (Capsicum annus L.) seeds are often slow and erratic, particularly under stress conditions. Field emergence trials sometimes have not responded to priming in pepper. This study examined the combined effects of matriconditioning and gibberellin application on the germination and stand establishment of pepper and tomato seeds. Ruminal degradability of hard or soft texture corn grain at three maturity stages Scientia Agricola

The predominance of vitreous endosperm in hard texture flint corn (Zea mays L.) can decrease ruminal starch digestion comparatively to the farinaceous endosperm of dent corn, reducing energy content of the grain. The objective of this experiment was to evaluate the effects of texture and maturity stage on ruminal degradability of corn grain. Two dent and two flint hybrids were harvested at the early dent, half milk line, and black layer stages. The proportion of vitreous endosperm (vitreousness) in dent hybrids was 44.3%, while in flint it was 67.0%. There was a linear increase in vitreousness with advancing maturity. Seed characters and their usefulness in the separation of Asteraceae species Biological Sciences

Studies on seed structure of Asteraceae have received little attention of botanists. Seed structure in Asteraceae is analyzed to evaluate the usefulness of characters in the separation of species through the analysis of nine weedy species. Seeds originate from anatropous, unitegmic and tenuinucellate ovules. The partial collapse of the developing seed coat is a common characteristic in the Asteraceae species studied. The testa consists of crushed and thin-walled cells in almost all of the species studied herein, except for Elephantopus mollis and Parthenium hysterophorus, which showed exotesta cells with U-shaped thickening. The analysis revealed high uniformity in seed characters.

It has been reported that Coffea arabica seeds contain as the main reserve protein, a legumin-like protein, constituted of two subunits, alpha and beta, of approximately 35 and 20 kDa. In this work the seed proteins of several coffee species and varieties were investigated by SDS-PAGE and gel filtration. No differences were observed in the electrophoretic profiles among varieties of C. arabica, however, marked differences were observed among species, or even among individuals of some species. In general, the molecular weight of the subunits alpha and beta accounted for a monomer of 48 to 62 kDa. The seeds of Caesalpinia echinata and C. ferrea behaved as typical endospermic seeds, despite their different morphological classification (exendospermic seeds were described for C. echinata and endospermic seeds for C. ferrea). Then, the aim of this work was to compare, under ultrastructural and histochemical terms, the nature of the storage substances and their accumulation sites, as well as the nutrient flow in seeds of these species. The effects of temperature on germination and endo-beta-mannanase activity in seeds of Sesamum indicum was investigated. The minimum germination temperature (Tmin) lies between 12.8°C and 13.2°C while the maximum temperature (Tmax) is located between 45.5°C and 46°C. Germinabilities are statistically not different from estimated viability (88%) between 18.8°C and 43.2°C. The Mann-Whitney test indicated the interval 31.9°C to 35.1°C as the optimum temperature (Topt) range for germination rate. The EBN is part of the post-zygotic hybridization barriers in the group and represents a reproductive isolating mechanism. Variability of ricin content in mature seeds of castor bean PAB

The objective of this work was to evaluate ricin concentration in castor bean seeds (Ricinus communis) of 20 accessions from the Banco de Germoplasma de Mamoneira of the Embrapa Algodão, Campina Grande, PB, Brazil, using the Enzyme Linked Immunosorbent Assay. Significant differences were observed among accessions. BRA 3271 had the highest ricin concentration in seeds (32.18 ng µg-1), and BRS Paraguaçu had the lowest (3.53 ng µg-1). There is the possibility of selecting genotypes with different ricin concentrations, which can be used according on the interest of the breeding programs.

Variation in zinc (Zn) concentration in the grain of 50 local rice varieties from Southern Thailand was examined by diphenyl thiocarbazone (DTZ) staining (intense, medium, low, and nil) and chemical analysis. Intense staining was found in the aleurone layer of 94% of the varieties, in the embryo 56% of the varieties and endosperm 12% of the varieties. Variation was found in the DTZ staining intensity of the embryo and endosperm of individual grains, with Shannon-weaver diversity index (H) of the DTZ staining at 0.15-1.00 for the embryo and 0.00-1.00 for the endosperm.
